Malyl-CoA lyase is an acetyl-CoA/glyoxylate-releasing C-C lyase in several central-carbon cycles. The enzymes from Rhodobacter species catalyze a step in the ethylmalonyl-CoA pathway for acetate assimilation; halophilic bacterial enzymes participate in the methylaspartate cycle in the malyl-CoA-forming direction; the enzyme from Chloroflexus aurantiacus participates in the 3-hydroxypropionate cycle for CO2 fixation. It cleaves (S)-malyl-CoA into acetyl-CoA and glyoxylate, and (2R,3S)-2-methylmalyl-CoA into propanoyl-CoA and glyoxylate. Structurally it is a malyl-CoA/beta-methylmalyl-CoA lyase of the citrate lyase family.
