Matriptase

Definition:

EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.21 Serine endopeptidases > 3.4.21.109 matriptase
Matriptase is a trypsin-like integral-membrane serine peptidase in family S1A implicated in breast-cancer invasion and metastasis; it can activate hepatocyte growth factor/scattering factor (HGF/SF) by cleavage of two-chain forms at an Arg to give active alpha- and beta-HGF (but does not activate plasminogen, which is homologous to HGF), and can also activate urokinase plasminogen activator (uPA), initiating the matrix-degrading peptidase cascade; hemojuvelin is a physiological substrate of matriptase-2; it cleaves synthetic substrates with Arg or Lys at P1 and prefers small side-chain residues such as Ala and Gly at P2. By activating HGF and uPA it drives epithelial carcinogenesis and matrix remodelling.
