Memapsin 1 is a membrane-anchored aspartic endopeptidase in peptidase family A1, atypical in carrying a C-terminal membrane-spanning domain. Although it can cleave beta-amyloid precursor protein to form the amyloidogenic beta-peptide implicated in Alzheimer's disease, it is not significantly expressed in human brain. It shows broad endopeptidase specificity and cleaves Glu-Val-Asn-Leu-/-Asp-Ala-Glu-Phe in the Swedish variant of amyloid precursor protein. In cells it is a gastrointestinal aspartic protease. It features a pepsin-like catalytic ectodomain and a single transmembrane helix.
