Memapsin 2 is the major beta-secretase proposed to cleave beta-amyloid precursor protein, generating the amyloidogenic beta-peptide implicated in Alzheimer's disease. It belongs to peptidase family A1 but is atypical in carrying a C-terminal membrane-spanning domain. It shows broad endopeptidase specificity and cleaves Glu-Val-Asn-Leu-/-Asp-Ala-Glu-Phe in the Swedish variant of amyloid precursor protein. In neurons it initiates amyloid plaque formation and is a leading Alzheimer's drug target. It features a pepsin-like catalytic ectodomain, a transmembrane anchor and a short cytoplasmic tail.
