Methylisocitrate lyase is an aldehyde-lyase of the methylcitrate cycle. It acts on threo-Ds-2-methylisocitrate but not on threo-Ds-isocitrate, threo-DL-isocitrate or erythro-Ls-isocitrate, cleaving (2S,3R)-3-hydroxybutane-1,2,3-tricarboxylate into succinate and pyruvate. It completes the propionyl-CoA assimilation cycle in bacteria and fungi. It is found in Eukaryota, Bacteria and Archaea. Structurally it is an isocitrate-lyase-superfamily TIM-barrel enzyme.
