Macrophage elastase is a zinc metalloendopeptidase in peptidase family M10 (interstitial collagenase family), synthesized as a 53-kDa proenzyme converted to a 22-kDa active form; cDNA sequences are known for mouse and human enzymes. It hydrolyzes soluble and insoluble elastin and also cleaves -Ala14-/-Leu- and -Tyr16-/-Leu- in the insulin B chain. In macrophages it degrades elastin and contributes to emphysema, aneurysm and matrix remodeling. It features an M10-family zinc catalytic domain.
