Pitrilysin is a zinc metalloendopeptidase from the periplasmic space of Escherichia coli, the type example of peptidase family M16, inhibited by EDTA and 1,10-phenanthroline but not thiol-dependent. It preferentially cleaves -Tyr16-/-Leu- and -Phe25-/-Tyr- bonds of oxidized insulin B chain and also acts on other substrates below 7 kDa such as insulin and glucagon. In the bacterial periplasm it digests small peptides. It features an M16-family zinc catalytic chamber.
