O-antigen ligase is a Gram-negative bacterial membrane enzyme that attaches the polymerized O-antigen molecule to the outer-core region of the lipid A-core oligosaccharide, finalizing lipopolysaccharide biosynthesis. Prior to the reaction both substrates are presented at the periplasmic face of the inner membrane, and the enzyme transfers the O antigen from its polyprenyl diphosphate membrane anchor onto the lipid A-core oligosaccharide, producing a lipopolysaccharide and releasing a polyisoprenyl diphosphate. It is required for assembly of the full-length LPS in the outer membrane.
