Papain is the type example of peptidase family C1, isolated from the latex of papaya fruit (Carica papaya), and inhibited by compound E-64 and proteins of the cystatin family. It hydrolyzes proteins with broad specificity for peptide bonds, but prefers an amino acid bearing a large hydrophobic side chain at P2, and does not accept Val at P1'. In organisms it serves as a broad-spectrum proteolytic defense and digestive enzyme, and is widely used in food, textile and biotechnological processing. It features the canonical papain-like catalytic cysteine-histidine-asparagine triad.
