Penicillopepsin is an aspartic endopeptidase from the imperfect fungus Penicillium janthinellum, with close relatives in P. roqueforti and P. duponti. It belongs to peptidase family A1. Like pepsin A it hydrolyzes proteins broadly, preferring hydrophobic residues at P1 and P1', but additionally cleaves Gly20-/-Glu in the insulin B chain; it clots milk and activates trypsinogen. In the fungus it digests extracellular proteins. It features a pepsin-like aspartic-protease fold.
