N-acetylglucosaminylphosphatidylinositol deacetylase is a membrane enzyme found in viruses and cellular organisms that catalyzes the second step of glycosylphosphatidylinositol (GPI) anchor formation in all eukaryotes. It appears to be a single subunit (PIG-L in mammals, GPI12 in yeast); in some species the sn-1 long-chain acyl of phosphatidyl is replaced by alkyl or alk-1-enyl. It hydrolyzes 6-(N-acetyl-alpha-D-glucosaminyl)-1-phosphatidyl-1D-myo-inositol into 6-(alpha-D-glucosaminyl)-1-phosphatidyl-1D-myo-inositol and acetate. In the ER it matures GPI anchors. It features a membrane deacetylase fold.
