PfpI endopeptidase is a cysteine endopeptidase from the hyperthermophilic archaeon Pyrococcus furiosus, the dominant protease in that organism. It efficiently cleaves Ala-Ala-Phe-/-7-amido-4-methylcoumarinamide. In the archaeon it contributes to intracellular protein turnover at near-boiling temperatures. It features a small thermostable cysteine-protease fold oligomerized into active multimers.
