Definition:

EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.23 Aspartic endopeptidases > 3.4.23.40 Phytepsin
Phytepsin is a plant aspartic endopeptidase best known from barley grain but present in other plants, belonging to peptidase family A1 but structurally atypical in carrying an internal ~100-amino-acid insertion not found elsewhere in the family. It prefers hydrophobic residues Phe, Val, Ile, Leu and Ala at P1 and P1', but also cleaves -Phe-/-Asp- and -Asp-/-Asp- bonds in 2S albumin from plant seeds. In seeds it mobilizes storage proteins during germination. It features a pepsin-like aspartic-protease fold with a plant-specific insert.
