Insulysin is an 110-kDa cytosolic zinc metalloendopeptidase known from mammals and the fruit fly Drosophila melanogaster, in peptidase family M16 (pitrilysin family). It is inhibited by bacitracin, EDTA, 1,10-phenanthroline and thiol-blocking reagents such as N-ethylmaleimide, but not by phosphoramidon. It degrades insulin, glucagon and other polypeptides but has no action on proteins. In cells it clears circulating insulin and peptide hormones. It features an M16-family zinc catalytic chamber.
