Proprotein convertase 1 (PC1/PC3, prohormone convertase 1) is a Ca2+-dependent subtilisin-like prohormone convertase in family S8, maximally active at pH ~5.5 and present in the regulated secretory pathway of neuroendocrine cells, acting cooperatively with PC2; substrates include pro-opiomelanocortin, prorenin, proenkephalin, prodynorphin, prosomatostatin and proinsulin, and unlike PC2 it does not hydrolyse pro-LH-RH, with prodynorphin processing unusually at a P2-Thr bond; it releases protein hormones, neuropeptides and renin from precursors generally by hydrolysis of Lys-Arg bonds. By maturing prohormones in dense-core vesicles it generates active peptide hormones. It appears in viruses and cellular organisms.
