Protein O-GlcNAcase
Definition:
EC Tree > 3 Hydrolases > 3.2 Glycosylases > 3.2.1 Glycosidases, i.e. enzymes that hydrolyse O- and S-glycosyl compounds > 3.2.1.169 protein O-GlcNAcase
Protein O-GlcNAcase is a dynamic post-translational modification-removing glycosidase that cleaves N-acetyl-beta-D-glucosamine from O-linked Ser/Thr residues of modified proteins, hydrolysing both [protein]-3-O-(N-acetyl-beta-D-glucosaminyl)-L-serine and the corresponding L-threonine conjugate into the free serine/threonine and N-acetyl-D-glucosamine. In higher eukaryotes O-GlcNAcylation is dynamic, inducible and abundant, regulating many cellular processes by interfering with protein phosphorylation; O-GlcNAc transferase (EC 2.4.1.255) adds the sugar and O-GlcNAcase removes it, making the enzyme a major therapeutic target in diabetes and neurodegeneration. It is distributed across Eukaryota and Bacteria.
