Thermolysin is a thermostable extracellular zinc metalloendopeptidase containing four calcium ions, the type example of peptidase family M4; possible species variants occur in Micrococcus caseolyticus and Aspergillus oryzae. Closely related but distinct enzymes include aeromonolysin, pseudolysin, bacillolysin, aureolysin and mycolysin. Its preferential cleavage is -/-Leu > -/-Phe. In thermophilic bacilli it digests extracellular proteins. It features a thermostable zinc catalytic fold stabilized by four Ca2+ ions.
