Pterocarpan synthase is a cytochrome-P450-dependent cyclase that forms the additional ring in pterocarpans, the basic phytoalexin skeleton of legumes, including (-)- and (+)-medicarpin and (-)- and (+)-maackiain. It requires the C-4 hydroxyl of the substrate to be in the (4R) configuration; the C-3 hydrogen configuration determines the final stereochemistry. Structurally it is a CYP93E pterocarpan synthase.
