Renin is a highly specific aspartic endopeptidase formed from prorenin in plasma and kidney, belonging to peptidase family A1 (pepsin A family) and found in viruses and cellular organisms. It cleaves the Leu-/-Val bond in angiotensinogen to generate the decapeptide angiotensin I, the first step of the renin-angiotensin-aldosterone cascade that controls blood pressure and sodium balance. In the kidney it is the rate-limiting enzyme of blood-pressure regulation. It features a pepsin-like aspartic-protease fold with a deep specificity cleft that recognizes the angiotensinogen N-terminal sequence.
