ADAM 17 endopeptidase is a membrane-anchored zinc metalloendopeptidase in peptidase family M12. In vivo it cleaves the membrane-bound 26-kDa tumor necrosis factor-alpha precursor at Pro-Leu-Ala-Gln-Ala-/-Val-Arg-Ser-Ser-Ser, releasing soluble 17-kDa TNF-alpha that induces inflammation, and similarly sheds other membrane-anchored cell-surface proteins. It shows narrow endopeptidase specificity. In cells it drives inflammatory cytokine release and ectodomain shedding. It features an M12-family zinc catalytic ectodomain linked to a disintegrin domain.
