Gametolysin is a zinc metalloendopeptidase in peptidase family M11, found in the periplasmic space of Chlamydomonas reinhardtii gametes as a 62-kDa inactive form that decreases to 60 kDa on activation. It is inhibited by phosphoramidon but thiol-activated. It cleaves the proline- and hydroxyproline-rich proteins of the Chlamydomonas cell wall, and also cleaves azocasein, gelatin and Leu-Trp-Met-/-Arg-Phe-Ala. During mating it digests the cell wall to allow gamete fusion. It features an M11-family zinc catalytic fold.
