Scytalidopepsin B is a second endopeptidase from Scytalidium lignicolum, insensitive to pepstatin and methyl 2-diazoacetamidohexanoate. 1,2-Epoxy-3-(p-nitrophenoxy)propane reacts with Glu53, which replaces one of the active-centre aspartates; the monomer, Mr 22,000, is one of the smallest aspartic endopeptidases. Similar inhibitor-resistant enzymes occur in Lentinus edodes, Ganoderma lucidum and Polyporus tulipiferae. It hydrolyzes proteins broadly, cleaving Phe24-/-Phe but not Leu15-Tyr and Phe25-Tyr in the insulin B chain. In the fungus it digests extracellular proteins. It features a monomeric aspartic-protease fold with a catalytic Glu.
