Staphopain is a broad-spectrum cysteine endopeptidase from Staphylococcus species and the type example of peptidase family C47. It acts broadly on proteins including elastin, but hydrolyzes small-molecule substrates rather limitedly; assays are conveniently performed with hemoglobin, casein or Z-Phe-Arg-NHMec. In the bacterium it contributes to immune evasion by degrading immunoglobulins and chemokines, and to tissue invasion. It features a catalytic papain-like fold secreted as an extracellular virulence factor.
