Ste24 endopeptidase
Definition:
EC Tree > 3 Hydrolases > 3.4 Acting on peptide bonds (peptidases) > 3.4.24 Metalloendopeptidases > 3.4.24.84 Ste24 endopeptidase
S2P endopeptidase is an integral membrane zinc metalloendopeptidase, the type example of peptidase family M50. The transcription factors SREBP-1 and SREBP-2 are synthesized as ER-membrane precursors and require two cleavages to release the active factor to the nucleus; this enzyme performs the second, site-2 cleavage. It cleaves type-2 transmembrane transcription factors within their membrane-spanning domains, known substrates including SREBP-1, SREBP-2 and forms of ATF6. SREBP-2 is cleaved at DRSR_ILL_483-/-CVLTFLCLSFNPLTSLLQWGGA, with the membrane-spanning segment underlined. In cells it couples cholesterol and ER-stress signals to transcription. It features an M50-family zinc catalytic site embedded in the membrane.
