Bontoxilysin is a zinc metalloendopeptidase produced by Clostridium botulinum, occurring as forms A-G differing in specificity on the neuroexocytosis apparatus, in peptidase family M27. The 150-kDa proenzymes are processed to disulfide-linked 100- and 50-kDa subunits, the latter carrying the endopeptidase activities, and are weakly inhibited by captopril and phosphoramidon. It performs limited hydrolysis of synaptobrevin (VAMP), SNAP-25 or syntaxin, with no action on small-molecule substrates. Acting at neuromuscular junctions, it blocks acetylcholine release and causes flaccid paralysis, in contrast to the spastic paralysis of tentoxilysin. It features an M27-family zinc catalytic light chain bound to a ganglioside-binding heavy chain.
