Thioglycine synthase requires Mg2+. It is found in anaerobic methanogenic and methanotrophic archaea, where it modifies a glycine residue in EC 2.8.4.1 coenzyme-B sulfoethylthiotransferase (methyl-coenzyme M reductase). Upon substrate binding, an external sulfide source attacks the target amide bond to form a tetrahedral intermediate; the amide oxyanion then attacks the γ-phosphate of ATP, releasing ADP and phosphate and yielding the thioamide. Structurally it is an ATP-dependent thiocarboxylate-forming enzyme.
