Trypanothione synthase
Definition:
EC Tree > 6 Ligases > 6.3 Forming carbon-nitrogen bonds > 6.3.1 Acid—ammonia (or amine) ligases (amide synthases) > 6.3.1.9 trypanothione synthase
Trypanothione synthase, characterized from several trypanosomatids such as Trypanosoma cruzi, catalyzes two successive condensations leading from glutathione and spermidine to N1,N8-bis(glutathionyl)spermidine (trypanothione): first glutathione plus spermidine plus ATP yields glutathionylspermidine plus ADP plus phosphate, then glutathione plus glutathionylspermidine plus ATP yields bis(glutathionyl)spermidine plus ADP plus phosphate. Some trypanosomatids (Crithidia, some Leishmania) contain only the first activity (EC 6.3.1.8). Structurally it is a trypanothione synthase (TryS).
