Ulp1 peptidase is a cysteine protease from Saccharomyces cerevisiae that recognizes human small ubiquitin-like modifier 1 (SUMO-1) as a substrate in both SUMO processing (alpha-linked peptide bonds) and SUMO deconjugation (epsilon-linked peptide bonds). Ulp1 has several functions, including an essential role in chromosome segregation and cell-cycle progression through G2/M. It hydrolyzes the alpha-linked Gly-Gly-/-Ala-Thr-Tyr bond at the C-terminal propeptide of the SUMO precursor Smt3, yielding mature SUMO, and also cleaves the epsilon-linked isopeptide bond between the C-terminal glycine of SUMO and a lysine side chain of substrate. In the nucleus it controls sumoylation dynamics. It features a cysteine protease catalytic domain related to the SENP family.
