XPF endonuclease

XPF endonuclease is a structure-specific 3'-flap endonuclease of the ERCC4/XPF family whose best substrates are a 3' flap followed by a nicked duplex, nicked three-way junctions and nicked four-way junctions; the enzyme from Sulfolobus is active only in the presence of the sliding clamp PCNA and cleaves the replication fork or 3'-flap structures in homodimeric form. By incising the DNA strand at the 3' side of lesion-containing bubbles it effects the dual-incision step of nucleotide-excision repair and contributes to DNA interstrand cross-link repair. It appears in viruses and cellular organisms and contains an ERCC4 nuclease domain plus a helix-hairpin-helix domain.
