C-8 Ceramide-1-phosphate
Based on 1 Customer Validation
C-8 Ceramide-1-phosphate ammonium is a short-chain synthetic C1P analog that inhibits apoptosis and increases cell survival. C-8 Ceramide-1-phosphate ammonium reduces MAPK activation and cytokine expression, induces Ca2+ mobilization and ERK1/2 phosphorylation, promotes M2-like polarization, and inhibits the M1 phenotype. C-8 Ceramide-1-phosphate ammonium inhibits acidic SMase, stimulates PI3-K/PIP3 generation, and interconverts with ceramide through kinase/phosphatase activity to maintain cellular homeostasis. C-8 Ceramide-1-phosphate ammonium can be used in research related to lipid metabolism and chronic wounds.

Description
In Vitro
C-8 Ceramide-1-phosphate (20 µM) ammonium inhibits the LTA+HA-driven inflammatory M1 program in human MDMs and reduces CD80 under inflammatory polarization[2].
C-8 Ceramide-1-phosphate (1-10 µM; 22 h) ammonium stimulates DNA synthesis in Rat-1 fibroblasts[4].
C-8 Ceramide-1-phosphate (5 µM; 48 h) ammonium increases the number of Rat-1 fibroblasts to 137% of the control, whereas C2-ceramide reverses this increase to 83%[4].
C-8 Ceramide-1-phosphate (10 µM; 10 min) ammonium does not decrease cAMP concentration in Rat-1 fibroblasts either with or without Forskolin (HY-15371)[4].
C-8 Ceramide-1-phosphate (50-150 µM) ammonium does not stimulate PLD in Rat-1 fibroblasts at mitogenic concentrations, and increases it 1.5-fold only at low concentrations[4].
C-8 Ceramide-1-phosphate (5 µM; 5 min) ammonium does not stimulate 42 or 44 kDa MAP kinase activity in Rat-1 fibroblasts[4].
C-8 Ceramide-1-phosphate (5 µM; 5 h) ammonium is metabolized to C8-ceramide in Rat-1 fibroblasts, with almost no conversion to S-1-P, sphingosine, or sphingomyelin[4].

-
Cell Line:Rat-1 fibroblasts
-
Concentration:1, 2, 5 and 10 µM
-
Incubation Time:22 h; [3H]thymidine during the last 6 h; effect maintained up to 28 h
-
Result:Produced about a 2.4-fold stimulation of [3H]thymidine incorporation into DNA after 22 h.
Maximum incorporation was obtained at 5 µM.
Concentrations higher than 10 µM were less effective in stimulating DNA synthesis or were inhibitory.
-
Cell Line:Rat-1 fibroblasts
-
Concentration:5 µM
-
Incubation Time:48 h
-
Result:Relative cell number with this compound (5 µM) was 137 after 48 h.
Relative cell number with this compound + C2-ceramide was 83.
C2-Ceramide completely reversed the effect of 5 µMthis compound on cell number.
