Biological Activity: Tubulin polymerization-IN-98 prodrug-1 is a prodrug prepared with the tubulin polymerization inhibitor A‑8d as the parent compound. Tubulin polymerization-IN-98 prodrug-1 binds to the colchicine binding site, and palladium-loaded resin catalyzes the efficient release of the active parent, thereby disrupting tubulin polymerization and dynamics. Tubulin polymerization-IN-98 prodrug-1 combined with palladium-loaded resin induces G2/M phase arrest, apoptosis, collapse of mitochondrial membrane potential, excessive ROS production, and inhibits endothelial cell migration and tube formation. Tubulin polymerization-IN-98 prodrug-1 combined with palladium-loaded resin achieves tumor growth inhibition in a HepG2 xenograft tumor-bearing mouse model. Tubulin polymerization-IN-98 prodrug-1 can be used for research related to hepatocellular carcinoma[1].
