f-MKKFRW acetate is a formylated MgrB signal sequence peptide and Fpr3 agonist. f-MKKFRW acetate can be produced by bacteria. f-MKKFRW acetate is selectively recognized by a subset of vomeronasal sensory neurons and activates Fpr3-expressing and B2m-expressing VSNs, but not NP2-expressing VSNs. f-MKKFRW acetate activates TRPM5-dependent signaling, triggering acetylcholine release. f-MKKFRW acetate drives innate avoidance behavior requiring Fpr3, Gαo, and Trpc2.

In Vitro
f-MKKFRW (8 min) acetate stimulates PTS in isolated mouse trachea through a chemosensory cell-dependent, TRPM5/PLCβ2/IP3R3-dependent, Atropine (HY-B1205)- and 4-DAMP-sensitive pathway[2].
f-MKKFRW (8 min) acetate increases the ciliary beating frequency in isolated mouse trachea from 8.7 Hz to 14.2 Hz[2].
f-MKKFRW (50 µM) acetate elicits transient Ca2+ elevation in a subset of TRPM5+ tracheal chemosensory cells (19/92; 21%)[2].
f-MKKFRW (10 µM; 15 min) acetate triggers TRPM5-dependent acetylcholine release in isolated mouse trachea, which can be detected by M3 reporter cells and HPLC[2].

Animal Model:C57BL/6N and transgenic lines (B6; ΔFpr3-lacZ, cGαo, Trpc2; adult male; 8-14 weeks old for behavioral tests; adult male for in-vivo VSN activation)[1]

Dosage:100 μM in PBS; 100 μM final concentration in 50% FEU; 100 μM final concentration in FEU; 50 μM final concentration in FEU

Administration:oronasal application; single exposure; free investigation/nasal contact; 10 min

Result:In freely behaving mice, both oronasal application and free investigation at 100 μM induced Egr1 expression in approximately 50% of Fpr3-positive VSNs; this activation was not observed with control stimuli.
In behavioral tests, 100 μM elicited robust avoidance in B6 mice, ΔFpr3-lacZ+/+, cGαo+/−, and Trpc2+/− mice.
Mice still displayed avoidance at 50 μM.
Avoidance was absent in ΔFpr3-lacZ−/−, cGαo−/−, and Trpc2−/− mice.
