Nav1.2-IN-4
Nav1.2-IN-4 is a blood-brain barrier-penetrant NaV1.2 inhibitor (IC50 = 31.21 μM). Nav1.2-IN-4 preferentially blocks the inactivated state of the neuronal voltage-gated sodium channel NaV1.2, inhibiting persistent and resurgent sodium currents associated with epileptogenesis. Nav1.2-IN-4 restores the GABA-glutamate balance by enhancing GABA and reducing glutamate. Nav1.2-IN-4 reduces oxidative stress in brain tissue by increasing GSH and decreasing MDA. Nav1.2-IN-4 protects mice against MES-induced tonic hindlimb extensor seizures and attenuates hippocampal neuronal degeneration in the PTZ kindling model. Nav1.2-IN-4 can be used for research on epilepsy.

Nav1.2 31.21 μM (IC50) |
In Vitro
Nav1.2-IN-4 exhibits high passive membrane permeability (6.53 × 10−6 cm/s)[1].
Nav1.2-IN-4 exhibits a favorable safety profile with no significant cytotoxicity up to 500 μM in HEK293T and SH-SY5Y cells[1].
Nav1.2-IN-4 (50 μM) shows low inhibitory activity against cardiac NaV1.5 channels (12.0% inhibition at 50 μM), indicating a low risk of NaV1.5-related cardiotoxicity[1].
Nav1.2-IN-4 exhibits minimal inhibition of major CYP subtypes (CYP3A4, CYP2D6, CYP2C9) at 50 μM[1].
Nav1.2-IN-4 is predicted to exhibit potent binding affinity for the NaV1.2 channel with a docking score of -9.1 kcal/mol, forming key hydrogen bonds and π-alkyl interactions within the binding pocket; molecular dynamics simulations from 10-100 ns indicate that this molecule forms a stable, energetically favorable complex with the NaV1.2 channel, with an MM-GBSA binding free energy of -32.3 kcal/mol, and the binding is primarily driven by van der Waals interactions.

In Vivo
Nav1.2-IN-4 (i.p.) shows an ED50 of 14.31 mg/kg and a protective index of > 34.94 in the MES model; and its LD50 is greater than 1000 mg/kg, indicating a favorable safety margin[1].
Nav1.2-IN-4 (30 mg/kg; i.p.; daily; 15 days) exerts the most significant and sustained protective effect against PTZ-induced kindling progression, and restores neurotransmitter balance and alleviates oxidative stress in the brain[1].
Nav1.2-IN-4 achieves brain-to-plasma ratios of 2.79 and 2.97 at 0.25 h and 0.5 h, respectively after intraperitoneal injection of 30 mg/kg, indicating significant brain exposure[1].
Nav1.2-IN-4 (30 mg/kg; i.p.; daily; 7 days) exhibits moderate but manageable hepatocellular stress, with a 74% increase in ALT[1].
