NF-κB-IN-21
NF-κB-IN-21 is a NF-κB signaling pathway inhibitor. NF-κB-IN-21 reduces the phosphorylation levels of IκBα and p65, and inhibits the nuclear translocation of p65. NF-κB-IN-21 downregulates the expression of iNOS, inhibits nitric oxide production, and decreases the levels of pro-inflammatory cytokines TNF-α and IL-6. NF-κB-IN-21 is used in the research of inflammatory diseases.

In Vitro
NF-κB-IN-21 (0.041-0.370 μM; 24 h co-incubation with LPS) inhibits LPS-induced activation of the NF-κB pathway in RAW264.7 cells in a dose-dependent manner by reducing the phosphorylation levels of IκBα and p65, and downregulates the expression of downstream iNOS[1].
NF-κB-IN-21 inhibits LPS-induced nuclear translocation of p65 in RAW264.7 cells, confirming its inhibitory effect on the activation of the NF-κB pathway[1].

Cell Line:LO2 human normal hepatocytes
-
Concentration:0.005-3.333 μM
-
Incubation Time:24 h, 72 h
-
Result:Showed no significant cytotoxicity across all tested concentrations after 24 h treatment.
Exhibited dose-dependent cytotoxicity with an IC50 value of 6.557×10-7 M after 72 h treatment, comparable to Celastrol's IC50 of 7.037×10-7 M.

Cell Line:RAW264.7 murine macrophage cells
-
Concentration:0.014-0.123 μM
-
Incubation Time:1 h pretreatment, followed by 24 h LPS co-incubation
-
Result:Dose-dependently inhibited LPS-induced IL-6 production, with near-complete inhibition at 0.123 μM, and significant inhibition at 0.041 μM.
Inhibited TNF-α production only at 0.123 μM, with no significant effect at 0.041 or 0.014 μM.
-
Cell Line:RAW264.7 murine macrophage cells
-
Concentration:0.041-0.370 μM
-
Incubation Time:24 h co-incubation with LPS
-
Result:Dose-dependently reduced LPS-induced phosphorylation of IκBα and p65, with significant inhibition observed at 0.123 μM and 0.041 μM.
Dose-dependently downregulated LPS-induced iNOS expression, consistent with NO inhibition results.
Left total p65 and IκBα levels unchanged across treatment groups.
