GLP-2 Receptor
The GLP-2 receptor (GLP-2R) is a class B GPCR that binds glucagon-like peptide-2, a proglucagon-derived peptide co-secreted with GLP-1 from intestinal L cells. Its expression is concentrated in the gut, where activation drives enterocyte proliferation, enhances nutrient absorption, tightens the mucosal barrier and increases intestinal blood flow. The clinically validated outcome is mucosal growth: the long-acting GLP-2 analog teduglutide activates GLP-2R to reduce parenteral nutrition dependence in short bowel syndrome, and related agonists are being tested in inflammatory bowel disease. Unlike its sibling GLP-1R, GLP-2R has little pancreatic expression and no direct insulinotropic action.
