Ubiquitin Conjugating Enzyme E2 V2

Encoded by the UBE2V2 gene and also known as Croc or UEV-2, this protein is the second human ubiquitin-conjugating enzyme variant (UEV). Like UEV1A, it lacks the active-site cysteine and cannot form ubiquitin thioesters; it functions instead by binding UBE2N (UBC13) as a non-catalytic partner, orienting the E2-ubiquitin intermediate so that K63-linked chains are assembled. UBE2V2/UBC13 complexes support NF-κB activation downstream of immune receptors and participate in DNA damage tolerance. UBE2V2 is broadly expressed across tissues and its complex with UBC13 is among the best-characterized K63 chain-forming machines. Recombinant UBE2V2 is used interchangeably with UEV1A to produce K63-linked polyubiquitin in vitro.
