Ubiquitin-Specific Protease 10

USP10 is a predominantly cytoplasmic deubiquitinase best known for deubiquitinating and stabilizing p53; in response to genotoxic stress, USP10 counteracts MDM2-mediated ubiquitination and promotes p53 accumulation. The enzyme also processes other targets, including the cystic fibrosis transmembrane conductance regulator (CFTR), and supports protein trafficking at endosomes. Through its interaction with G3BP1, USP10 additionally participates in stress granule dynamics. USP10 activity is attenuated in certain tumors with wild-type p53, while its expression supports tumor cell survival in others, presenting context-dependent biology. Its G3BP1-binding motif and catalytic core have both been explored as starting points for inhibitor discovery.
