Ubiquitin-Specific Protease 6

USP6, also known as TRE17 or Tre-2, is a deubiquitinase with low expression in most adult tissues that became prominent as a proto-oncogene. Its activation occurs through chromosomal rearrangements, most famously the COL1A1-USP6 and CDH11-USP6 fusions that drive aneurysmal bone cyst, as well as fusions found in nodular fasciitis and related myofibroblastic tumors. These rearrangements place USP6 under control of highly active promoters, and the oncoprotein signals through NF-κB, Wnt/β-catenin and matrix metalloproteinase pathways to promote proliferation and invasion. Recombinant USP6 supports studies of its unusual N-terminal TBC-type region and of its transforming mechanisms.
