Ptaquiloside
Based on 1 Customer Validation
Ptaquiloside (Braxin C; Aquilide A) is an illudane-type sesquiterpene glycoside natural toxin derived from ferns, with carcinogenic properties. Ptaquiloside can be converted to Pterosin B (HY-N1570). Ptaquiloside upregulates the expression of metallothioneins Mt1 and Mt2, and depletes intracellular free zinc ions. Ptaquiloside downregulates the expression of CD44 and CD107a in lesion-infiltrating CD8+ T cells, impairs T cell activation and degranulation capacity, and promotes immune evasion. Ptaquiloside can be used in cancer-related research.

In Vitro
Ptaquiloside (10-60 μg/mL; 24 h) activates the ATR-Chk1 DNA damage response in AGS cells, increases γH2AX, and maintains high P53 protein levels, with Chk1 phosphorylated at Ser317 at 60 μg/mL[3].
Ptaquiloside (10-60 μg/mL; 2-24 h) transiently increases P53 transcript levels at 2-6 h and decreases P53 mRNA at 24 h in AGS gastric epithelial cells[3].
Ptaquiloside (60 μg/mL; 24 h) downregulates DNA damage response and DNA repair genes in AGS cells, increasing GADD45A, SESN1, XPC, and PCNA, and decreasing DDIT3, MAP2K6, NBN, RAD50, and RAD51L1[3].
Ptaquiloside-derived PTQ dienone reacts with double-stranded DNA (ODN-3/ODN-4) to form O6-PTBdG and N7-PTBdG guanine adducts[4].
