In Vitro
SARM1 activator 1 induces time-dependent changes in NAD+ and cADPR levels in HeLa-SARM1-HA-xFlag cells[1].
SARM1 activator 1 (25-50 μM; 24 h) alters axon morphology in DRG neurons of wild-type mice, and at 50 μM (48 h) alters axon morphology in DRG neurons of SARM1 knockout mice[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:HeLa cells ectopically expressing full-length SARM1 (HeLa-SARM1-HA-3×Flag and HeLa-SARM1-HA-xFlag) and HeLa cells
-
Concentration:10 μM (screening); different concentrations (ATP-level measurements)
-
Incubation Time:24 h (screening)
-
Result:Among 1.36 million compounds, 7-(4-ethoxyphenyl)-3-((pyridin-3-ylmethyl)thio)-6,7-dihydro-5H-imidazo[2,1-c][1,2,4]triazole was identified as a superior SARM1 activator and named G10/Compound 004.
IC50 was ≤4 μM.
Cell survival ATP level (%) was shown for different G10 concentrations in HeLa-SARM1-HA-xFlag and HeLa cells.
