SCR-7952
Based on 1 Customer Validation
SCR-7952 is an orally active MAT2A allosteric inhibitor (IC50 = 18.7 nM; Ki = 14.49 nM; Kd = 0.56 nM). SCR-7952 reduces SAM production, PRMT5 activity, and SDMA marks by binding to the MAT2A allosteric pocket, and dysregulates RNA splicing through FANCA intron retention, thereby selectively inhibiting the proliferation of MTAP-deleted cancer cells and producing synergistic effects with PRMT5 inhibitors. SCR-7952 can be used for research on MTAP-deleted cancers.

SCR-7952 (0.3 mg/kg; p.o.; once daily) combined with Onametostat (JNJ-64619178) (HY-101564) produces near-complete tumor stasis in the HCT116 MTAP-/- xenograft model, with a TGI of 94.4%[1].

-
Animal Model:Nu/Nu (female)[1]
-
Dosage:0.25, 0.5, 1.0, or 3.0 mg/kg
-
Administration:p.o. (oral gavage); once daily (q.d.)
-
Result:Compared with the vehicle group, oral administration of SCR-7952 led to a significant dose-dependent reduction in tumor growth.
Tumor growth inhibition (TGI) was 60.0% at 0.5 mg/kg, 62.7% at 1.0 mg/kg, and 82.9% at 3.0 mg/kg.
In a satellite group, SAM levels in HCT116 MTAP-/- xenograft tumors and mouse plasma were significantly decreased, with 53.56-68.72% inhibition at 2 or 4 h post final dose in the low group.
-
Animal Model:Nu/Nu (female)[1]
-
Dosage:0.3 mg/kg
-
Administration:p.o. (oral gavage); once daily (q.d.)
-
Result:Modest efficacy was observed with either SCR-7952 (0.3 mg/kg) or JNJ-64619178 (1.0 mg/kg) alone.
Almost complete tumor stasis was achieved when both agents were combined, significantly better than monotherapies (TGI 94.4% vs. 55.0% vs. 54.0%).
SCR-7952 (0.3 mg/kg) or JNJ-64619178 (1.0 mg/kg) alone both led to a remarkable reduction of SDMA levels.
In the combination group, a significant reduction of SDMA levels was observed compared with single-agent groups.

In Vivo:
Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: 0.5 mg/mL (1.43 mM); Clear solution; Need ultrasonic
This protocol yields a clear solution of 0.5 mg/mL.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (5.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
