PF-670462 is a selective CK1δ and CK1δε inhibitor (Ki of 10 nM and 50 nM, respectively). PF-670462 is also a ZDHHC8-specific inhibitor (Kd 15.9 μM). PF-670462 inhibits CK1-mediated LRRK2 phosphorylation. PF-670462 blocks PER phosphorylation and CK1δ/ε-dependent TGF-β signaling. PF-670462 reduces GPX4 palmitoylation, sensitizing tumor cells to Ferroptosis inducers. PF-670462 can be used for research on Parkinson's disease, NRAS-mutant melanoma, pulmonary fibrosis, chronic lymphocytic leukemia, diabetic nephropathy, and melanoma .
