PVtCD-cTP-AlkF-2 is a ROS-responsive DAPK1 BmTAC activator with blood-brain barrier permeability. PVtCD-cTP-AlkF-2 is equipped with a VHL E3 ubiquitin ligase ligand modified with a ROS-responsive phenylboronic acid caging group, and undergoes uncaging activation only in a high-ROS microenvironment. PVtCD-cTP-AlkF-2 can selectively recognize and degrade dephosphorylated activated DAPK1. PVtCD-cTP-AlkF-2 exerts its activator function through caveolae-mediated transcytosis. PVtCD-cTP-AlkF-2 reduces APP and Tau phosphorylation, decreases Aβ oligomer deposition, and improves cognitive function in aged APP/PS1 mice. PVtCD-cTP-AlkF-2 can be used for research on Alzheimer's disease .

Cou-Coxib is a ROS-responsive fluorescent Celecoxib (HY-14398) prodrug that is activated by ROS triggering and self-immolation to release Celecoxib and the fluorophore Cou-OH. Celecoxib released from Cou-Coxib elevates intracellular ROS levels, induces S-phase arrest in MDA-MB-231 triple-negative breast cancer cells, and downregulates the expression of apoptosis-related proteins Bcl2, Survivin, and procaspase-3. In the H9C2 cardiomyoblast model, Cou-Coxib attenuates the cardiotoxicity caused by Celecoxib, maintains mitochondrial membrane potential, upregulates IL-10, and exhibits biocompatibility in developing zebrafish embryos. Cou-Coxib can be used for research on triple-negative breast cancer .

IRS1 is an integrin-targeted, ROS-responsive self-assembling peptide prodrug molecule with selective anti-cancer activity against integrin-overexpressing tumor cells. IRS1 contains an RGD motif for integrin binding, can covalently bind to DR4/DR5, and promotes DR4/DR5 aggregation. After oxidation by ROS, IRS1 undergoes a morphological transition from nanoparticles to nanofibers, exposes the pharmacophore of Chlorambucil (HY-13593), disrupts cell membrane integrity, activates the extrinsic apoptosis pathway, and can penetrate and inhibit the three-dimensional uveal melanoma spheroid model. IRS1 can be used for the research of uveal melanoma .
