Xylocydine is an inhibitor of CDK1/2, inhibiting CDK function in various apoptotic processes. Xylocydine specifically blocks CDK1 and CDK2 activity with IC50 of 1.4 nM and 61 nM, respectively. In addition, Xylocydine downregulates phosphorylated nucleolin and retinoblastoma protein levels. .

BRD4/AKT-IN-1 is a BRD4/AKT inhibitor with BRD4 IC50 66.12 nM and AKT1 IC50 143.81 nM. BRD4/AKT-IN-1 blocks BRD4-mediated c-Myc transcriptional regulation, modulates AKT1 signaling, decouples AKT phosphorylation from pro-survival effectors. BRD4/AKT-IN-1 induces G0/G1 cell cycle arrest via downregulated phosphorylated RB, cyclin E1, CDK2. BRD4/AKT-IN-1 elevates LC3B levels to promote autophagy. BRD4/AKT-IN-1 promotes apoptosis in cancer cells. BRD4/AKT-IN-1 can be used for the research of metastatic castration-resistant prostate cancer .

TMLB-G9 is an orally active B3GAT3 inhibitor (human Kd = 0.72 μM). TMLB-G9 blocks glycosaminoglycan biosynthesis and inhibits chondroitin sulfate biosynthesis in hepatocellular carcinoma cells. TMLB-G9 induces apoptosis by upregulating cleaved PARP and cleaved caspase-3. TMLB-G9 induces G0/G1 cell cycle arrest by upregulating Rb and downregulating CDK1, CDK2, Cyclin A2, Cyclin E1, and phosphorylated Rb. TMLB-G9 inhibits migration and invasion by reversing epithelial-mesenchymal transition, manifested as upregulation of E-cadherin and downregulation of N-cadherin, vimentin, MMP-2, MMP-9, and ZEB1. TMLB-G9 inhibits colony formation. TMLB-G9 exhibits broad-spectrum antitumor activity against multiple cancer cell lines. TMLB-G9 can be used for research on hepatocellular carcinoma .
