PROTAC stands for proteolysis-targeting chimeras, which are heterobifunctional small-molecule compounds containing two different ligands: one is a ubiquitin ligase E3 ligand, and the other is a ligand that binds to the target protein in cells. The two ligands are connected by a linker, thereby forming a "ternary" polymer-target protein ligand-Linker-E3 ligand. PROTACs specifically degrade target proteins through the "ubiquitin-proteasome" pathway by bringing the target protein and intracellular E3 ubiquitin ligase into closer proximity. PROTACs can improve the druggability of "undruggable" targets.
