6-PhEt-ATP-γ-S
6-PhEt-ATP-γ-S (N6-(Phenethyl)-ATPγS) is a bulky ATP analog probe carrying a thiophosphate group. 6-PhEt-ATP-γ-S, as an orthogonal phosphate donor, is specifically utilized by mutant-sensitive kinases such as CDK12-AS and Slt2-as to label their direct substrate proteins. 6-PhEt-ATP-γ-S specifically binds to the enlarged pocket of mutant kinases via its bulky side chain (avoiding wild-type), and covalently labels substrates through thiophosphorylation. 6-PhEt-ATP-γ-S can be used for kinase-substrate profiling studies.

Biological Activity
Description
In Vitro
6-PhEt-ATP-γ-S (1 mM; 30 min) supports CDK12-AS/CCNK-mediated thiophosphorylation of recombinant LEO1 in a cell-free assay[1].
6-PhEt-ATP-γ-S (10 mM; 30 min) is specifically utilized by Slt2-as kinase in a cell-free system (an in vitro kinase system containing yeast extract or E. coli recombinant protein) to transfer the thiophosphate group to substrate proteins (such as Rlm1, Msg5, Rcn2, Gga1, Caf20, etc.)[2].
