AIM4
Based on 1 Customer Validation
AIM4 is a TDP-43 aggregation inhibitor. AIM4 binds to the TDP-43 C-terminal amyloidogenic core and C-terminal fragments, antagonizing liquid-liquid phase separation, amyloid-like aggregation, and oligomer formation, while polarizing microglia toward an anti-inflammatory state. AIM4 can be used for research on amyotrophic lateral sclerosis.

Biological Activity
Description
In Vitro
AIM4 (50-200 μM; 4 h) reduces the formation of TDP-43-YFP punctate aggregates in S. cerevisiae erg6Δ cells, decreasing from approximately 20% to approximately 10% after 4 h under 0.01% galactose conditions, and from approximately 50% to approximately 25% under 0.1% galactose conditions[1].
AIM4 (200 μM; 2 h) dissolves preformed TDP-43-YFP punctate aggregates in S. cerevisiae erg6Δ cells, reducing the proportion of cells containing aggregates from 35.5% to 22.56%[1].
AIM4 (200 μM; 4 h) does not alter TDP-43-YFP expression levels or yeast viability under these conditions, although yeast expressing TDP-43-YFP grows relatively faster in the presence of AIM4[1].
AIM4 (1-2 mM; 1 h) is internalized by BV2 microglial cells, but its uptake is lower than that of ABA and ABE[2].
AIM4 (50 mM stock solution in 25% DMSO; protein:AIM4 1:15; 37°C overnight) completely inhibits the amyloid aggregation of recombinant TDP-432C-A315T[3].
AIM4 (50 mM stock solution in 25% DMSO; protein:AIM4 1:15; 5 h) inhibits the in vitro liquid-liquid phase separation of recombinant TDP-432C-A315T[3].
AIM4 (6 mM; 15 h) completely inhibits recombinant TDP-432C aggregation in the ThT fluorescence assay and eliminates the early acceleration of TDP-432C aggregation induced by HSA Protein (HY-P1956)[4].
AIM4 is a nuclear protein with a functional NLS that may act as a negative regulator of the Slt2 and Hog1 pathways in Saccharomyces cerevisiae, and aim4∆/aim4∆ cells exhibit increased resistance to copper, 1,3,7-trimethylxanthine, and zymolyase[5].
