Antascomicin B
Antascomicin B is a macrocyclic natural product that binds purified human FKBP12, FKBP12.6, FKBP51, and FKBP52. Antascomicin B acts as a molecular glue that binds the FK1 domain to stabilize the FKBP51-Akt1 complex, with a Kd of 6.7 nM for human FKBP51, independent of AKT activation status and Hsp90 binding.

In Vitro
Antascomicin B binds the FK1 domain of human FKBP51 and forms additional hydrogen-bond contacts with G84 and a solvent-exposed variable region[1].
Antascomicin B stabilizes the FKBP51-Akt1 complex in HEK293T cells, enhances FKBP51 photocrosslinking and Akt1 co-immunoprecipitation, and does so independently of Hsp90 binding and AKT T308/S473 activation status[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
