Coumarin-based kinetic stabilizer-2
Coumarin-based kinetic stabilizer-2 is a 3-substituted coumarin-carbamate-distal substructure derivative and a kinetic stabilizer of amyloidogenic immunoglobulin light chain (FL LC) dimers. Coumarin-based kinetic stabilizer-2 stabilizes the native conformation of immunoglobulin light chains, reduces protein misfolding and proteolytic hydrolysis, with an EC50 value of 77.1 nM for the human target and an EC50 value of 27.9 nM for the WIL-FL T46L/F49Y variant. Coumarin-based kinetic stabilizer-2 can be used in research related to immunoglobulin light chain amyloidosis.

Biological Activity
Description
In Vitro
Coumarin-based kinetic stabilizer-2 binds the JTO-FL LC dimer at 2.0 Å resolution (PDB 7LMN), its carbamate linker forms hydrogen bonds with F98 and V96, and its phenyl-imidazole distal group is located near Y49′[1].
