Coumarin-based kinetic stabilizer-3
Coumarin-based kinetic stabilizer-3 (compound (R)-83) is a full-length immunoglobulin light chain (FL LC) kinetic stabilizer and a derivative of coumarin. Coumarin-based kinetic stabilizer-3 binds to and stabilizes the amyloidogenic WIL-FL T46L/F49Y dimer (EC50 < 2.5 nM) and the native state of WIL-FL (EC50 = 4.1 nM), slows misfolding, proteolysis, and aggregation, and antagonizes amyloidogenic unfolding. Coumarin-based kinetic stabilizer-3 can be used in research on immunoglobulin light chain (AL) amyloidosis.

Biological Activity
Description
In Vitro
Coumarin-based kinetic stabilizer-3 (10 μM; 1 h) kinetically stabilizes the WIL-FL T46L/F49Y/C214S light chain dimer, conferring a 0.63-fold protection against Proteinase K hydrolysis[1].
